Macrophages have established roles in tumor growth and metastasis, but information about their role lung promotion is limited. To assess the of macrophages tumorigenesis, we developed a method minimally invasive, long-term macrophage depletion by repetitive intratracheal instillation liposomal clodronate. Compared with controls treated doses PBS-containing liposomes, resulted marked reduction number size at 4 mo after single i.p. injection carcinogen urethane. After urethane treatment, increased M1 marker expression during first 2–3 wk, followed M2 week 6. Using strategy to reduce alveolar initiation early stages (weeks 1–2) or late progression 4–16), found significantly fewer smaller tumors both groups compared controls. Late-stage reduced VEGF impaired vascular tumors. In contrast, early-stage urethane-induced NF-κB activation lungs development premalignant atypical adenomatous hyperplasia lesions 6 wk injection. Together, these studies elucidate an important for indicate that cells distinct different carcinogenesis.
